II. Report back to-does from last meeting TO-DO list from last week:

1. Yeast Strain - finish registration on ATCC (Credit refs) 2. Next meeting at BioCurious: have a planned experiment ready for the meeting; contact Nicola to coordinate details 3. Contact sequentech about sequencing reactions and primer design - 4. Modify experiment timeline google doc: add additional sheets to overall document, one for each month, to break monthly calendar schedule into a week-by-week schedule for precise tuning of progress

IIIa Stretch goals Mammoth Cheese - problem - unecc glamourous, not taken seriously Bioreactor - 1 large - need might not be understood by general public, but sounds cool Bioreactor - several small Micelle equipment: particle analyzer Endangered - black-footed ferret - too glam but may raise awareness of endangered species... but might be too much new info which muddies/decreases understanding of both projects

Molecular Biology Experiments

Run SDS-PAGE gels on K-casein from E. coli Order primers to verify construct Send out K-casein for sequencing Make competent yeast Transform yeast Run SDS-PAGE gels on K-casein from yeast

Things we need:

   - Uracil
   - Reagents to induce competence
   - Buy gels or pour our own? BUY! We gots $
   - SDSbuffer, loading dye and Molecular Weight marker.

Cheese experiment 2

Tenatively scheduled for the Omni 4799 Shattuck Ave July 28 7PM

Can we dissolve casein powder, and make cheese out of it?

Stuff to bring:

  • Matts high-shear blender
  • jewelry cleaning sonicator (patrik will bring)
  • Do we have lab-sonicators? (may be more adjustable)
  • heater (juul will verify if omni has stoves)
  • pH meter (juul will order one, lou will check home depot and aquariam store)
  • chymosin (juul will bring)
  • white vinegar (lou will bring)
  • thermometer (digital ?) (juul & lou will bring)
  • cheese cloth (Patrik will bring)
  • pot (lou will bring)
  • CaCl (juul will bring)
  • test tubes / glasswares
  • ice (lou will bring)

David Poole agreed to bring the following. 1- micellar casein powder (oddly missing from our list) 2- whole milk (control) 3- fat free milk 4- milk-free fat 5- citric acid 6- extra calcium salt 7- extra rennin/chymosin 8- extra jewelry sonicator 9- papers on prior work from the journal of dairy science 10- additional stuff I've found useful for making cheese (e.g. a salad spinner, thermometer etc)

Parameters to vary: pH, Ca, T, sonication/shear test using chymosin, and settling over time

ToDo:

  • Collect all papers relevant to experiment, and give access to everyone (juul)
  • Solicit questions for ask a biohacker and tell us why we're wrong (juul)
  • Research protein purification methods (juul)
  • Organize / set up inventory
