Remaining iGEM material due by midnight Friday Oct 17! Judging form due (Teams should highlight their best BioBrick Parts, and indicate which special prizes each team thinks they should be eligible for.) Giant Jamboree wiki FREEZE at 11:59PM EDT We need to port EVERYTHING to the iGEM wiki! Additional Part DNA due to the Registry (not required, however parts are not eligible for medals and awards) Do we want to use one of the iGEM templates? http://2014.igem.org/WikitemplateB_home or http://2014.igem.org/WikitemplateA_home Template B.by general consensus and for ease of use. 3. Yeast expression What were the results of all our swabbing experiments last week? Did anyone actually record the outcome? 5-FOA - did anyone order it? Craig Ordered (and paid -$67.43- please reimburse from zymoresearch, 100x solution; 10/13/14- details added in the online ledger) How does it work? http://www.zymoresearch.com/buffers-solutions/chemicals/5-fluoroorotic-acid-5-foa in strains expressing the functional URA3 gene coding for orotine-5-monophosphate decarboxylase that is involved in the synthesis of uracil. Yeast strains that are phenotypically Ura+ become Ura- and 5-FOA(R) (resistant) after selection because 5FOA is toxic to cells - yeast with a functional Ura3 gene die in the presence of 5-FOA. Next steps for yeast work? Grow up yeast, use 5-FOA to kill Ura3+ cells.

5. Question for Johan & any other BioCurians re: human kappa (kex -). I know that the transformation 0f the 9/12/2014 cloning reaction yielded only 2 colonies once and 0 colonies on a repeat, but have you repeated the Electra cloning for human kappa (kex -)? Might be an easy fix. They have done a new Electra reaction. Grew at room temperature. Few small colonies that didn't grow in LB carb. Bad Electra enzyme after multiple freeze thaws??
