Monday Nov 17th

Repeat Mini Prep of bovine alpha S1 and bone beta B yeast:

Participants: Lafia, Meenakshi, Nikola, Johan

Materials: 1 Zymo yeast mini prep kit 2 tubes of transformed bovine Alpha S1 2 tubes of transformed bovine Beta B 1 agarose gel electrophoresis with gelgreen instead of EtBr

Method:

The Zymo protocol was deviated from. For step one, we spun down a pellet from 10ml of liquid culture. The pellet was spun down at 1200 rpm for 6 minutes. Following this we followed Zymo protocol from step 2 exactly.


Result: 3 bright bands were observed, they appeared to be around the 10kb mark of the Zymo DNA ladder. There was a missing band for one bovine Alpha S1.

Result Explanation: The transformation is likely to have worked. The plasmid was not linearized, so that may explain why it was near the 10 kb marker rather than 6 kb (the size of the plasmid).
